General Warnings
  • Malignancy

    This medicine may increase the risk of lymphoma and other malignancies. Doctors should inform patients about these risks and advise the patient to prevent exposure to sunlight by wearing protective clothes.

  • Serious infections

    The chances of developing infections are higher in the patients receiving this medicine. Hold the treatment and start with broad-spectrum antibiotics if there are any signs and symptoms of fever. Close monitoring of the body temperature and the blood cell count is necessary.

  • Kidney impairment

    This medicine is not recommended in the patients with a history of kidney impairment. Close monitoring of kidney function is necessary. The dose should be adjusted based on the creatinine clearance (CrCl).

  • Liver impairment

    This medicine is not recommended in patients with a history of liver impairment. The concentration of this medicine may increase in such patients. The baseline of the liver function tests should be obtained before starting the treatment.

  • Hyperkalemia

    This medicine may increase the levels of potassium in the body and may increase the risk of arrhythmia. Close monitoring of potassium levels is necessary. Medicines that can increase potassium levels like ACE inhibitors and potassium-sparing diuretics should be avoided.

  • Hypertension

    Use with caution in hypertensive patients. Appropriate antihypertensives should be administered to control the blood pressure. Do not use medicine that may increase the potassium levels.

what are the adverse effects of Gramune capsule 100 mg 50’s?

Hyperlipidaemia, hyper-uricaemia, hyperkalaemia, hypomagnesaemia, tremor, headache, paraesthesia, hypertension (discontinue if cannot control), GI upset, anorexia, abdominal pain, gingival hypertrophy, hepatic and renal dysfunction, hypertrichosis, muscle cramps, myalgia, fatigue, anaemia, thrombocytopenia, signs of encephalopathy, rash, oedema, weigh gain, malignancies, Iymphoproliferative disorders, infections. Rarely, micro-angiopathic Haemolytic anaemia or haemolytic uraemic syndrome, menstrual disturbances, gynaecomastia, hyperglycaemia, motor polyneuropathy, pancreatitis, muscle weakness, myopathy, optic disc oedema. Risk of benign intracranial hypertension; investigate patients presenting with signs of raised intracranial pressure, withdraw cyclosporin if diagnosed.

What happens if I miss a dose of Gramune capsule 100 mg 50’s?

Organ transplantations: Used alone, Adults & Children over 3 mth: 10-15 mg/kg by mouth 4-12 yr before transplantations followed by 10-15 mg/kg daily for 1-2 wk postoperatively then reduced gradually to 2-6 mg/kg daily for maintenance (dose should be adjusted according to blood-cyclosporin concentration and renal function); dose lower if given concomitantly with other immunosuppressant therapy; if necessary one-third corresponding oral dose can be given by IV infusion over 2-6 hr.
Bone marrow transplantation; prophylaxis and treatment of graft versus-host disease: Adults & Children over 3 mth: 3-5 mg/kg daily by IV infusion over 2-6 hr from day before transplantation to 2 wk postoperatively (or 12.5-15 mg/kg daily by mouth) then 12.5 mg/kg daily by mouth for 3-6 mth then tailed off (may take up to a yr after transplantation).
Nephrotic syndrome: Adults: Orally, 5 mg/kg daily in 2 divided doses. Children: 6 mg/kg daily in 2 divided doses.
Atopic dermatitis: Adults: Orally, initially 2.5 mg/kg daily in 2 divided doses, if good initial response not achieved within 2 wk. Increase rapidly to max 5 mg/kg daily; initial dose of 5 mg/kg daily in 2 divided doses if very severe. Children: Not recommended.
Severe psoriasis: Adults: Orally, initially 2.5 mg/kg daily in 2 divided doses, increase gradually to max 5 mg/kg daily if no improvement within 1 mth (discontinue if response still insufficient after 6 wk); initial dose of 5 mg/kg daily justified if condition requires rapid improvement. Children: Not recommended.
Severe arthritis: Adults: Orally, initially 2.5 mg/kg daily in 2 divided doses, if necessary increased gradually after 6 wk; max 4 mg/kg daily (discontinue if response insufficient after 3 mth); dose adjusted according to response for maintenance and treatment reviewed after 6 mth. Children: Not recommended.
